A shop is a chest which has been configured to buy and/or sell an item at a specific price. A chest can only buy/sell a single specific item.
Buying & Selling
To buy and sell from a shop, hit the shop sign, and then follow the on-screen prompts. For a chest that offers both buy and sell features, hit the sign of the chest that you are interested in.
Setup
To create a shop chest, place a chest on the ground, hit it with the type of item you wish to buy/sell, and then follow the on-screen prompts. A double-chest is required to both buy & sell.
